Background: There appears to be a close interrelationship among myosin phenotype, size, number of myonuclei and myonuclear domain in skeletal muscle fiber in a variety of mammals. However, with regard to this interrelationship little data are available for the mouse. Objective: The aim of the present study was to examine the relationship among the type of myosin heavy chain (MHC) expressed, the cross sectional area (CSA), the number of myonuclei and the myonuclear domain in single fibers of slow-twitch soleus muscles and the fast-twitch tibialis anterior and muscles of adult mice. Experimental design: Muscle fibers of adult C57BL mice were analysed by SDS-PAGE for their MHC expression in isolated soleus muscle fiber (SMF) and tibialis anterior muscle fiber (TAMF), and by confocal microscopy used to count their myonuclear numbers, and to estimate cytoplasmic volume/myonucleus (myonuclear domain). Results: The analysis of MHC revealed that SMF contained the largest population of MHC I fibers. On the other hand, the analysis of MHC revealed that TAMF contained the largest percentage of MHC IIb fibers. The myonuclear number in both type IId/x and IIa fibers in TAMF was significantly greater than that of type IIb. The myonuclear domain of the type IIb in TAMF was significantly greater than in either type IId/x or IIa. There were significant positive correlations between myonuclear domain and CSA in SMF and TAMF for 0.549 and 0.857, respectively. Conclusions: The myonuclear domain seems to be correlated with greater absolute size of muscle fiber. These larger domains in predominantly fast muscles may reflect the lower metabolic rate than that which occurs in predominantly slow muscle. The data suggests that the different type muscles are not only unique in their MHC composition but also have relatively distinct physiological and biochemical properties. These data also provide a baseline for future studies of myonuclear plasticity in the mouse model.
