Coexpression of HLA-DR and Melanoma-Associated Antigens and Effects of Gamma Interferon on Human Melanoma Cell Lines

Expression of HLA-DR and melanoma-associated antigens (MAA) was investigated in six human melanoma cell lines using two-color flow cytometry, monoclonal antibodies and recombinant human γ-interferon. Monoclonal antibodies used for the study were, 96.5, 225.28S and L243, which recognize 97 kd MAA p97, high molecular weight (HMW)-MAA, and HLA-DR respectively. Cells were first stained with 96.5 or 225.28S, then stained with L243, and the dual-stained cells were analyzed using a fluorescence-activated cell sorter (FACS). All six melanoma cell lines examined expressed these antigens to various degrees (HLA-DR : 7〜100%, p97 : 45〜100%, HMW-MAA : 33〜73%) without γ-interferon treatment. In the cell line A101D, p97 (+) cells were always HLA-DR (+). In contrast, HLA-DR (+) cells were always p97 (+) in three other cell lines KHm-3/ps, KHm-3 and KHm-6. Almost all cells coexpressed HLA-DR and p97 in the remaining two cell lines KHm-1/4 and KHm-1/w. As concerns HMW-MAA, HMW-MAA (+) cells were always HLA-DR (+) in two cell lines A101D and KHm-1/4, and HLA-DR (+) cells were always HMW-MAA (+) in the cell line KHm-6. In the cell line KHm-1/w, most HMW-MAA (+) cells coexpressed HLA-DR. After γ-interferon treatment (500IU/ml, 72 h), an increase of HLA-DR (+) cells was observed in one cell line KHm-6, and an increase of p97 (+) cells was noted in the cell line KHm-3. On the other hand, γ-interferon treatment decreased p97 (+) cells in two cell lines KHm-6 and A101D, and HMW-MAA (+) cells in four cell lines KHm-6, A101D, KHm-1/4 and KHm-1/w. Two-color flow cytometric analysis showed that there was a strong correlation between the expression of these two antigens, i. e., the HLA-DR (+) melanoma cells tended to express MAA, and vice versa. These results suggest that γ-interferon and the expression of these antigens on tumor cells play important roles in the antitumor host reactions.
