The present study demonstrated that two species of cells from mice infected with S. enteritidis attenuated SER strain showed a cytotoxic activity against L 929 cells. The results obtained are as follows. 1) Peritoneal exudative cells (PEC) from Balb/c (H-2^d) mice infected with salmonella markedly inhibited ^3H-TdR incorporation by L 929 (H-2^k) cells. In a parallel experiment also, these activated PEC showed a significant cytotoxicity against L 929 cells, which was measured by ^<51>Cr-release from the target cells. These activities became apparent in 7 days after infection and persisted for at least 46 days. 2) PEC from Balb/c mice pretreated with zymosan as well as with picibanil also inhibited ^3H-TdR incorporation by L 929 cells. At the same time, these activated PEC were found to be cytotoxic against L 929 cells. 3) Spleen cells from Balb/c, C57BL/6 (H-2^b) and C3H/He (H-2^k) mice infected with salmonella showed the cytotoxic activity against normal as well as salmonella-infected L 929 cells on the 7th and 11th days after infection. The immunological reactivity, however, was insignificant on the 14th day. 4) In contrast, spleen cells from C3H/He mice, but neither from Balb/c nor C57BL/6 mice, infected with Sendai virus or vaccinia virus both showed the cytotoxic activity against L 929 cells on 5th day after infection but the immunological activity of these cells declined to low level by the 12th and 14th days. The same cytotoxicity of the immune cells, however, appeared against virus-infected L 929 cells, but not against uninfected cells. In conclusion, in the viral infections, cytotoxicity appeared only when immune spleen cells and target cells share the H-2 haplotype; on the contrary, in the present experiments of salmonellosis, this H-2 compatibility requirement was found apparently unnecessary.
