A New Malondiamide Synthesis in Liquid Ammonia Alkylation of Malondiamide with Alkali Hydroxide

Alkylation of malondiamide was achieved by the reaction between the active methylene group of malondiamide and alkyl halide after using alkali hydroxide in liquid ammonia as metalation reagent. The process was named as malondiamide synthesis with alkali hydroxide in liquid ammonia. As alkali hydroxides, lithium, sodium and potassium hydroxides were used. And benzylation, ethylation, propylation, and butylation of malondiamide were performed in liquid ammonia at room temperature, the yeild being 70〜85 per cent. For explanating the speciality of the new synthetic method, the authors have proposed the following hypothesis. Alkali hydroxide in liquid ammonia is in equilibrium with alkali amide and produced water is fixed by liquid ammonia as ammonium hydroxide. The following reaction formulas were assumed : NaOH(in NH_3) + NH_3 〓 NaNH_2 + H_2O(in NH_3) H_2O(in NH_3) + NH_3 →NH_4OH CH_2 〓CONH_2 CONH_2 + NaNH_2 〓 [CH〓CONH_2 CONH_2]^- Na^+ + NH_3 [CH〓CONH_2 CONH_2]^- + RX →CHR〓CONH_2 CONH_2 + X^-
