H^+-ATPase inhibitor, bafilomycin A1, reduces bone resorption during experimental movement of rat molars

This study was designed to clarify the effects of administering a specific inhibitor of vacuolar-type H^+-ATPase, bafilomycin A1, on the bone resorbing activity of osteoclasts during experimental movement of rat molars. To generate orthodontic force by the Waldo method, an orthodontic elastic band was inserted between the upper first and second molars for 7 or 14 days. After elastic band application, bafilomycin A1 solution (10^<-7>M or 10^<-5>M) was administered daily intraperitoneally (10μl/day/rat). Control rats received the same volume of physiological saline. The extents of molar movement and the structural changes of osteoclasts were examined by means of light, and both scanning and transmission electron, microscopy. In control rats, elastic band insertion increased the mean interdental space between the first and second molars to approximately 307μm on day 7 and to 363μm on day 14. Bafilomycin A1 administration significantly and dose-dependently reduced the mean interdental space on both days, presumably by blocking the osteoclastic resorption of surrounding alveolar bones. In bafilomycin A1-administered rats, most osteoclasts on the alveolar bone surfaces had lost their intact ruffled borders and associated cytoplasmic vacuoles, the functional site of bone resorption. These results suggest, for the first time, that bafilomycin A1 administration effectively reduces the extent of experimental movement of rat molars by inhibiting osteoclastic bone resorption via a mechanism involving impairment of the structure and function of osteoclast-ruffled borders.
