ラット正中口蓋縫合軟骨の成長, 分化における圧縮力の影響

正中口蓋縫合軟骨は両側上顎骨の間に存在し, その生力学的特徴から二次軟骨であると考えられている.この研究はラット正中口蓋縫合軟骨の, 骨あるいは軟骨分化への圧縮力の影響を明らかにすることを目的とする.4週齢のウィスター系雄性ラットの正中口蓋縫合軟骨に, 歯科矯正用ワイヤーを用い, 両側第一臼歯を介して1, 2, 4, 7, あるいは14日間, 種々の大きさの外力を負荷した.正中口蓋縫合軟骨の分化過程はI型, II型コラーゲンを用い免疫組織化学的手法により検索した.組織学的, 免疫組織化学的に, 対照群のラットの正中口蓋縫合軟骨は二次軟骨の特性を示していた.実験群では未分化間葉系細胞を含む正中口蓋縫合軟骨の中央部において, 成熟軟骨細胞への分化がみられ, II型コラーゲンの免疫組織化学的反応は実験期間の増加とともに増大した.この反応は正中口蓋縫合軟骨に負荷された外力の大きさと実験期間の長さに関連しており, 外力の増大により軟骨分化はより進行した.この結果は, 正中口蓋縫合軟骨の前軟骨細胞層に存在する, 未分化間葉系細胞の成熟軟骨細胞への分化が圧縮力により制御されているということを示唆している.
