Transmembrane Chondroitin Sulfate Proteoglycans in the Developing Brain : Involvements in Signal Transduction as Well as in Cell Adhesion

NG2 proteoglycan(NG2-PG), recepto-type protein tyrosine phosphatase ζ / β(RPTPζ / β), and neuroglycan C(NGC)are the major transmembrane proteoglycans bearing chondroitin sulfate chains in the developing mammalian brain. NG2-PG, expressed on glial progenitor cells, binds to several extracellular matrix constituents including type VI collagen, tenascin and laminin. This proteoglycan is co-localized with platelet derived growth factor(PDGF)α-receptor on the progenitor cells, and the interaction between these two molecules is required for the optimal response of the cells to PDGF. RPTPζ / β binds to pleiotrophin, a mitogenic and neurite-promoting factor, in addition to some adhesion proteins such as Ll / Ng-CAM, N-CAM and tenascin. From these findings, it is postulated that transmembrane chondroitin sulfate proteoglycans are involved not only in the cell adhesion but also in the signal transduction of growth factors. In the developing rat brain, we recently found NGC, a neuronal proteoglycan with an epidermal growth factor(EGF)-like module in its ectodomain and some phosphorylation sites in both the ectodomain and the cytoplasmic domain. The experimental data, though limited at present, suggest that NGC also participates in transmembrane signaling as well as in cell adhesion during rat and chicken brain development.
