Characterization of normal and photoaged cutaneous microfibrils

Dermal elastic fibers consist of amorphous elastin and microfibils. Microfibrils are present throughout elastic fiber elements, including elaunin and oxytalan fibnls. Fibrillin molecules compose the structural backbone of cutaneous microfibrils. We have found that versican and LTBP-1 interact with fibrillin-1 and localize to cutaneous microfibrils. Thus, by binding to fibrillin microfibrils, these two ligands can impart additional functional properties to cutaneous microfibrils. In photoaged dermis, aggregation of elastin and microfibrillar components (solar elastosis) is characteristically observed. The functional and morphological properties of versican and LTBP-1 in photoaged microfibrils (microfibril aggregates) were analyzed by various histochemical and biochemical approaches. From these results, we conclude that alteration of microfibrils in solar elastosis is responsible for the phenotypical changes of skin aging.
