Meth A腫瘍の腹腔内増殖過程におけるサイトカイン誘導一酸化窒素合成の抑制(免疫学)

多くの種類の腫瘍細胞内で一酸化窒素ラジカル(NO・)が合成される.我々は,腹腔内でのMeth A線維肉腫増殖過程におけるNO・産生を経時的に測定した.腹水中のマクロファージ数が増加し,腫瘍移植3-6日で誘導性NO合成酵素(iNOS)量および活性は最高となった.LPSおよびIFNγ刺激に対する細胞の反応性は,9および11日で顕著に低下した.腫瘍細胞の増殖は,NO・産生と反比例した.電子常磁性共鳴解析およびニトロタイロシン免疫染色では,NO・産生にともなって形成される特徴的な細胞集族像は検出されなかった.これらの結果から,NO・産生はMeth A腫瘍の増殖とともに抑制されると結論される.NO・産生の低下はTGFβ,IL-10の抑制性サイトカイン量とは関連しなかったが,セピアプテリン(テトラハイドロバイオプテリンのプロドラッグ)の添加により産生はある程度回復した.このように,腫瘍の増殖過程において,iNOSの機能発現に必須な共因子の抑制は,NO・産生の抑制に関与すると思われる.
