Accumulation of the Products of β-Conglycinin α'-Subunit Gene Constitutively Expressed in Seeds and Non-Seed Tissues of the Transgenic Petunia Plants

Transgenic petunia plants which harbored a chimeric gene consisting of the promoter sequence from cauliflower mosaic virus (CaMV) 35S transcript and the coding sequence and 3' end sequence of the α'-subunit gene of soybean β-conglycinin led to accumulation of the gene products in all tissues examined. The amount of α'-mRNA was equivalent in seeds and leaves, but seeds accumulated approximately 30-fold greater levels of α'-protein than leaves because stability of the α'-protein was presumable different in each tissue. The intact α'-subunit (76 kDa) and a 55 kDa breakdown polypeptide of the α'-subunit accumulated in seeds of the transgenic plants. By contrast a polypeptide of 80 kDa, the predicted size of the α-precursor, and a 53 kDa breakdown polypeptide were found in leaves and other non-seed tissues. Both seed type α-proteins (76 and 55 kDa) and non-seed type α'-proteins (80 and 53 kDa) reacted with the antibody raised against β-conglycinin. Subcellular fractionation experiments showed that the non-seed type α-proteins were localized in microsomal membrane fractions in leaves. Seed type α'-proteins accumulated only in seeds during mid- to late-stages of seed maturation, while non-seed α'-proteins were found in non-seed tissues and in seeds of early maturation stage.
