Activity of Key Enzymes Involved in Carbohydrate Metabolism in Phaseolus vulgaris Cell Suspension Cultures

Activities of some key enzymes of glycolysis and sucrose metabolism were investigated in relation to the physiological growth stage in bean cell suspension cultures. Activities of sucrose synthase, pyrophosphate:fructose-6-phosphate phosphotransferase, ATP:fructose-6-phosphate phosphotransferase, UDP glucose pyrophosphorylase, acid and alkaline invertase were detected. Both pyrophosphate:fructose-6-phosphate phosphotransferase and sucrose synthase activities increased during the active phase of cell division. Thereafter activities began to decline when sugar in the medium was depleted. The increase in enzyme activities coincided with a sharp decrease in the endogenous sucrose, glucose and fructose levels. The largest change occurred in the activity of sucrose synthase, which was more than seven fold higher in logarithmic phase cells than in lag phase cells. Transfer of mid-logarithmic phase cells to fresh medium, containing 93 mmol dm^<-3> sucrose, or addition of sucrose to existing medium, resulted in a further increase in PP_i:fructose-6-phosphate phosphotransferase and sucrose synthase activities.
