The Effect of Oxygen Radicals on Proteolysis in Isolated Oat Chloroplasts

When chloroplasts isolated from oat leaves were incubated in the light, proteolysis and the activity of an acid endopeptidase increased. The increases were suppressed by 5 mM mannitol, an hydroxyl radical scavenger. The proteolysis and the endopeptidase also increased when chloroplasts were treated with Asc-Fe system, which can generate hydroxyl radical. In the polypeptides contained in chloroplasts, the hydrolysis of seven polypeptides was promoted by the treatment with Asc-Fe. Results suggest that oxygen radicals might mediate the light- and oxygen-induced proteolysis.
