Amplification of Extrachromosomal Circular DNA in Intact Bean Leaves Treated with Benzyladenine

Bean (Phaseolus vulgaris L.) leaves were treated with benzyladenine (BA) beginning 7 days after sowing, and were harvested at 21 days. There were less than 10 molecules per cell of small polydisperse circular (spc) DNA in mesophyll cells from untreated leaves, and more than 30 molecules in the BA-treated cells. The spcDNA ranged in size from 0.2 to 7.5 μm, with a mean length of 1.7 μm.
