Photosynthetic CO_2-fixing activity in dark-grown spruce seedlings

Bicarbonate-dependent O_2-evolving activity in dark-grown cotyledons of Picea abies was measured with an oxygen electrode with different preillumination times. The activity showed a slight linear increase with increasing preillumination time. On the other hand, O_2-evolving activity (Hill activity) of chloroplasts prepared from preilluminated dark-grown cotyledons exhibited a characteristic change of a steep rise followed by a gradual increase with increasing preillumination time. The results obtained were discussed in connection with the light activation of the latent, inactive O_2-evolving center in dark-grown cotyledons.
