Glutamate and glutamine were produced when intact proplastids of in vitro cultured tobacco cells were incubated with nitrite and α-ketoglutarate. ATP stimulated the production to a considerable extent. While glutamate was the main product of incubation in the complete medium, glutamine production surpassed glutamate production when proplastids were incubated without α-ketoglutarate. These facts suggests the operation of the glutamine synthetase/glutamate synthetase pathway in the pro-plastids. This suggestion was substantiated by the demonstration of both glutamine synthetase and glutamate synthetase activities in the proplastid fraction.
