Effects of manganese, calcium, dithiothreitol and bovine serum albumin on the light-reactivation of Tris-acetone-washed chloroplasts

Oxygen-evolution activity of spinach chloroplasts was investigated by washing chloroplasts with 0.8 M Tris buffer containing 20% acetone. This inactivation was easily removed by two successive treatments, dark- and light-reactivations. The first treatment was a dark-reactivation step, rewashing inactivated chloroplasts with reduced DPIP (DPIP treatment). The second one was a light-reactivation step, incubating chloroplasts with Mn^<2+>, Ca^>2+>, dithiothreitol and bovine serum albumin under illumination. Both light- and dark-reactivation treatments were required to regain oxygen-evolution activity of Tris-acetone-washed chloroplasts, which is characteristic of such chloroplasts. However, in Tris-washed chloroplasts considerable activity was recovered by dark-reactivation alone. Manganese and calcium contents of Tris-acetone-washed chloroplasts were compared with those of chloroplasts obtained by other preparations. Tris-acetone washing was presumed to inhibit the oxygen-evolution site of Photo-system II by affecting Mn, Ca and other substances in chloroplasts. The inhibition site was estimated from a change in fluorescence yield of chlorophyll and the effect of artificial electron donor specific for Photosystem II on NADP photoreduction activity.
