A good yield of structurally intact chloroplasts isolated from wheat for electron microscope examination was obtained with the application of glutaraldehyde fixative as a primary macerating and isolating medium. It was not always necessary to take special precautions in tonicity or to hesitate to store the chloroplast suspension for delayed electron microscopic preparation. Some noticeable features found in senescent chloroplasts are described.
