A study was made on the chemical kinetics of the hydroxyl ton catalyzed hydrolysis of distigmine bromide (1-methyl-3-hydroxypyridinium bromide-1,6-hexamethylene-bis-N-methylcarbamate) in solution to evaluate its stability in ophthalmic solution. Mechanism of this hydrolysis was found to consist of consecutive first-order reactions by thinlayer chromatography and ion-exchange chromatography. Analog computation of ultraviolet (UV) spectral data and quantitative ion-exchange chromatography were used to determine the rate constants of the consecutive reaction. The temperature dependency of each reaction was established, and the Arrhenius parameters were determined.
