Electroencephalographic (EEG) and behavioral effects of atropine and scopolamine were examined and also the effect of atropine and scopolamine on the convulsion induced by nikethamide, pentetrazol and acetylcholine in cats, dogs, rabbits, guinea-pigs, rats, and mice. 1) Atropine and scopolamine (0.1-10 mg/kg) caused, in all the species except guineapigs, bursts of 15-16 cps spindle waves or high-voltage slow (1-2 cps) waves in the cortical and subcortical EEG. 2) In guinea-pigs, atropine (higher than 100 mg/kg) had no effect on their EEG, but scopolamine in 1-5 mg/kg produced high-voltage slow waves. 3) Nikethamide in 80-100 mg/kg induced convulsion on behavior and spike or seizure pattern on EEG in all species except guinea-pigs. Pretreatment (10 min) of the animals with atropine antagonized the convulsion and the spike of seizure pattern induced by nikethamide, but pretreatment with scopolamine did not have such an antagonistic effect. 4) Pentetrazol in 20 mg/kg induced convulsion on behavior and spike or seizure pattern on EEG in rabbits and cats. Neither atropine nor scopolamine had no effect on the Pentetrazol convulsion. 5) Intracisternal injection of acetylcholine (1.0 μg/0.1 ml) into curarized cats produced a spike pattern on EEG. Both atropine and scoploamine antagonized this spike induced by acetylcholine. 6) Consequently, the difference between atropine and scopolamine is found only in their antagonistic effect on the convulsion induced by nikethamide.
