In order to examine the action of various chemicals on the crystalline lens, a new experimental method was devised using the crystalline lens in vitro. The conditions of the lens after excision was examined on the basis of whether the cationic transport was normal or not. Changes in the condition of the lens would result in its opacity and also by various biochemical changes. In the present experiment, cationic transport function of the lens decreased first, and outflow of potassium from the lens was found to precede other changes. In the in vitro experiment, outflow of potassium from the lens was accelerated by ice cooling and by the administration of ouabain or monoiodoacetic acid. These evidences suggest that the examination of cationic transport of the lens by the present method is an effective means in the examination of lens functions.
