Following administration of ^3H-2-(7-methoxy-10-methyl-2-phenothiazinyl)-propionic acid (PRT) into gastrointestinal tract of male rats by the ligated loop method, most of the radioactivity was found to be readily absorbed from the intestine. After oral administration of ^3H-PRT into male and female rats, the radioactivity level in blood increased rapidly and reached a maximum at 30 min. ^3H-PRT level in the blood was in proportion to the dosage between 8 and 200mg/kg. Most of ^3H-PRT in the blood was bound to plasma proteins. The radioactivity distributed in each organ reached a maximum within the first 1 hr after the administration of ^3H-PRT and gradually decreased thereafter in both male and female rats. In particular, high level of radioactivity was found in the liver, kidneys, and gastrointestinal tract. After the administration of ^3H-PRT, approximately 85% of the administered radioactivity was excreted into the bile within 24 hr, while 90% or more of the administered radioactivity was excreted in the urine and feces within 96 hr. Any significant difference was not found in the excretion of radioactivity between male and female rats. ^3H-PRT was given to male rats after daily administration of PRT for 30 days. No difference was found in the behavior of ^3H-PRT between the rats pretreated with PRT and those untreated.
