Digitonin solution of cattle thodopsin and opsin was spectroscopically examined over the ragne of wave length from 210mμ to 600mμ. The absorption spectra obtained immediately after preparation are presented in Fig. 1. Besides A, B and C-band, there is found a shoulder near 230mμ in both the spectra of rhodopsin and opsin. This will be provisionally called D-band. The ultraviolet absorption of rhodopsin, as shown in Fig. 2, changes gradually with time after preparation (aging effect), giving anyhow a difference spectrum with two peaks at 225mμ and 260-270mμ (Fig. 3), as levealed similarly in the case of mild heat denaturation of opsin (Fig. 5). As ionic strength, digitonin concentration or hydrogen ion concentration becomes higher, the aging effect mentioned above seems to be more and more suppressed (Fig. 4). With regard to the ultraviolet absorption of rhodopsin and opsin, it is supposed that tyrosine and tryptophan may be responsible for D-band as well as C-band.
