The present report deals with the structural constiuents interphase macronucleus of Paramecium caudatum, i. e. nuclear membrane, karyolymph, nucleolus-like bodies and chromatin. The observations on the morphological properties of nuclear membrane and karyolymph was not beyond the resolving power of the ordinary light microscope. The nucleolus-like bodies, which have basophilic nature and lose their affinity for pyronine by the action of ribonuclease or by the treatment with cold perchloric acid solution, can be divided into three groups as A, B, and C. It seems that they have RNA-rich nature in common. The members of group A are somewhat granular in appearance and are 1 to 3μ in diameter. Sometimes they aggulutinate in a form of a rosary or a ring, and the number of them is in the level of a hundred usually. From their constancy as to the macronuclear constituents and their reactivity to the cytochemical treatments, group A can be considered as nucleolus. The member of group B, which can be occasionally observed in the macronucleus of old material in decayed culture medium, lacks their constancy as a constituent and shows correlative changes associated with the physiological condition of material. They are hardly been detected in the rapidly mass-cultured one. The nncleolus-like bodies of group C, granular and about 2μ in diameter, showa a curious affinity to dyes. The peripheral region of this body has an affinity to Feulgen's staining, but the central zone shows pyroninophilic and Feulgen negative. Though this structure is designated as nucleolus-like body for the convenience of description, it is a few constituents of macronucleus and is recongnizable constantly. Chromatin may be divided into two groups, A and B. They are Feulgen positive and are believed to have DNA-rich nature as a result of the action of desoxyribonuclease, hot trichloro-acetic acid and molar NaCl solution. Chromatin A is one of the constant constituents of macronucleus and shows various shapes such as granule, rod, and fibre. It is probable that the variability of shape may be the property of chromatin A. In some cases, the fibrous one of this group in interphase is 2 to 5μ in length. Chromatin B which has strong affinity to Feulgen's staining is assumed to the unstable structure and show relative changes of the number and size by the physiological conditions of the animal. Constituents such as chromatin and nucleolus-like bodies are also observed in the interphase macronucleus of P. aurelia, P. bursaria and Tetrahymena geleii.
