The present paper deals with the results of morphological observations on the development of the macronuclear anlage in the exconjugant of Paramecium caudatum. Paramecia studied were from stocks KA6 and K_32 belonging to types XXIII and XXIV of variety 12, respectively. The results are summerized as follows. 1. In the course of the macronuclear development are distinguished four successive stages-formation of heterochromatic aggregates, development of nucleoli, enlargement of the anlage, and increasing of basopilia in the anlage. Through thses stages, four of the eight micronuclei develop into the vegetative macronuclei. 2. In the early stage of development, the micronucleus is swollen. Some chromosomes in the four nuclei, remaining partly condensed, agglutinate one another, thus forming about ten heterochromatic aggregates, each consisting of paired, heterochromatic portions, which are deeply stained with basic dyes. Reddish purple metachromasia could be seen by staining with toluidine blue after acetic alcohol fixation. A large number of paired chromatin threads which are lightly stained with basic dyes are observed around the aggrates. 3. Al large contral and then many, scattering small nucleoli make their appearance in association with the heterochromatic aggregates which are on the way of disintegration. 4. After the nucleli are formed, the anlages enlarge rapidly, and consequently the Feulgen stainability is markedly reduced. The nuclear structure in this stage is rather clearly observed in acetic orcein squash preparations, in which deeply stained granules are scattered, corresponding to the optical sections of uncoiled chromosomes. 5. After the enlargement, the anlage gradually becomes more basophilic. In different stages of further macronuclear development, we can see two sorts of structure, one with the paired thinner and the other with the scattered thicker threads, each probably representing the two alternating, duplication stages of chromosomes. It seems that the duplication of chromosomes in the anlage takes place under fairly uncoiled conditions like that of the macronuclear chromosomes in the vegetative phase.
