1. The melanophores which had expanded in vivo under definite conditions were unable to be contracted by KCl solution after isolation. 2. The melanophores pretreated with glucose solution could expand in KCl solution regardless of treatment with NaCl, KCl or NaCl-KCl mixture solution before glucose-treatment. The ones which had expanded in KCl solution, when transferred into glucose solution, showed contraction or the temporal contraction followed by re-expansion. 3. The melanophores pretreated with glucose solution could contract in KCl solution after having immersed in Ringer solution for one minuute. The pretreatment with NaCl solution seems to be necessary for the occurrence of contraciton in KCl solution. On the other hand, when the NaCl concentration of the solution for treatment was lower than that of NaCl solution for pretreatment, the melanophores caused contraction. From these results it seems highly probable that the contraction produced by transferring from NaCl into KCl solution is not due to KCl but the leakage of NaCl from the melanophores. 4. The melanophores pretreated with glucose solution could expand in solutions of NH_4Cl, BaCl_2, CaCl_2, KBr and fructose and the ones pretreated with Ringer solution contracted in those solutions, as similarly as in KCl solution. 5. The reactions of the melanophores produced by transferring from the one solution into the other might be explained on an assumption that the solutions examined have the effect of expanding the melanophores and the velocities of their penetration into and cut of the melanophores vary with the concentration of the solutes.
