1.The critical volume-ratio (n=Na:K)of the mixture of NaCl and KCl solutions at the concentration of M/7.5, which is capable of expanding the melanophores having contracted without any previous treatment, in isolated scales of Oryzias latipes is about 1.3. 2.The critical value of n of the mixture which is able to contract the melanophores having beforehand expanded in Ringer solution, is about 7, and generally the contraction observed here is transient and followed by re-expansion. 3.The facts that the K concentration of the solution which is capable of barely expanding the melanophores having contracted in vivo, is higher than that of contracting the melanophores having expanded in Ringer solution, and that the Na concentration of the former is lower than that of the latter, led us to the following assumption: The plasma membrane of melanophore is permeable to those cations and the expansion by Na and the contraction by K are produced by the difference in the concentration of each ion between the internal environment of melanophore and the external medium in which the melanophore is immersed.
