The granules found in the cytoplasm of the so-called primordial germ cells have been regarded as one of the prominent characteristics of the primordial germ cells in amphibians. In the present study, the nature of these granules was examined histochemically. In the larvae of Hynobius nebulosus employed in this study, the granules are observed during a period from 30 days up to 50 days after egg-laying. The distribution and amount of granules change with the differentiation of primordial germ cells, and the granules decrease in number and in size as the transformation of nucleus and the absorption of yolk-plateletes proceed. The granules assume black-brown colour when stained with Delafield-haematoxylin and eosin, however, these were almost completely decoloured when treated with Cl_2 and H_2O_2, and showed negative reactions for both Feulgen reagent and Safranine reagent after Kaufman, McDonald and Gay's method. Similar granules are discernible in the embryonic cells of developing intestinal epithelium, and after aforsaid histochemical examinations these showed the very same reactions as that observed in the case of primordial germ cells. Considering on the facts obtained it was concluded that the granules in primordial germ cells were melanine-like substance produced generally in the developing embryonic cells as a result of cellular metabolism, and therefore, it was incorrect to regard these granules as an essential morphologic determiner of the primordial germ cells.
