This paper deals with the activation (breakdown of the cortical alveoli and formation of the perivitelline space) of Fundulus eggs in relation to the extracellular medium, and with their electrical properties (membrane potential and membrane resistance) during activation. The "auto-activation" of Fundulus eggs immersed in normal sea water without insemination was investigated by changing the concentration of sea water and glucose solution. Activation took place in normal sea water (or 1 M glucose solution), and in hypertonic sea water (2 M glucose solution), but not in hypotonic sea water (weaker than 50% sea water). These facts indicated that Fundulus eggs were not really auto-activated, but that the activation was caused merely by hypertonicity of the extracellular medium. Electrical experiments were performed in 30% sea water and in normal sea water. Activation was induced by an electrical stimulus of 0.5 to 1×10^<-7> A in order to insert a micro-electrode through the ooplasmic membrane. Membrane potentials of unactivated eggs and activated eggs were a few mV negative with respect to 30% sea water and in normal sea water. In both media, a slow hyperpolarization (activation potential) was observed during activation. Membrane resistance decreased during activation and reverted back to the original value after completion of activation.
