New methods for injecting fluid and for transferring cytoplasm between individuals of Paramecium are described. Needles are made of Pyrex tubes by the aid of a microelectrode puller. The tip of needles is broken off by pushing them gently against a piece of paper to make the inside diameter of 2-3μ (for fluid-injection) or about 5μ (for cytoplasm transfer). The needle, held in the holder of micromanipulator, is connected by plastic tubing to a 50μ liter syringe controlled by a micrometer screw. The whole system from the needle tip to the syringe is completely filled with mineral oil. Recipient, or donor and recipient paramecia are delivered into a hanging drop in a moist chamber with open front. The drop should be small enough to restrict the motion of paramecia by surface tension. The needle is moved almost horizontally, but the tip is introduced at an angle of about 10°. Calibration of the amount of injected material is accomplished in the following way. A small amount of a dilute detergent solution is drawn into the oil-filled needle, and a minute amount of the oil is similarly drawn into the needle. After measuring the length of the short column of the oil with an ocular micrometer, the oil is expelled into the dilute detergent solution. Calibration is completed by calculating the volume of the resulting spherical oil droplet from its measured diameter.
