Preparation and Characterization of Vitellogenin-like Protein from the Serum of an Estrogen-Injected Japanese Quail

Estrogen administration to the male or immature female of Coturnix induced the systhesis of two new kinds of proteins, vitellogenin-like protein and low density protein, both of which normally occur in the blood serum of the egg-laying female. The vitellogenin-like protein (VLP) can be fairly well isolated from the serum by chromatography on DEAE-Sepharose without any degradation in the presence of a protease inhibitor, phenylmethylsulfonyl fluoride (PMSF). Employing SDS- and non-SDS-polyacrylamide gel electrophoresis, together with the immunodiffusion test, it was determined that Coturnix VLP may occur as two molecules with a minor difference in molecules weight, in other words, as a doublet molecule. Molecular weights were estimated to be approximately 240, 000 and 220, 000 respectively. This VLP was further split into five polypeptide bands on a SDS-polyacrylamide gel by incubation at 37℃ for 24 hrs without PMSF, while incubation with PMSF did not permit such splitting.
