The rapid freeze substitution method has proven to be an excellent preserver or morphology at the electron microscopical level. Since, with the use of acrolein or glutaraldehyde as fixative , this method has recently yielded results in the enzyme histochemistry of alkaline phosphatase and acid phosphatase in kidney tissues(Saito and Takizawa, 1986). We have here applied it to retinal tissues for the precise lotalization of the cyclic nucleotide metabolizing enzymes.
