With enzyme solutions extracted from wheat, barley and millet sprouts, general properties of nucleases were studies and nucleotides obtained from RNA hydrolysates were identified.1) The optimun pH values of nucleases and phosphatases from these sprouts were found in both acidic (pH 4.5-5.0) and alkaline (pH 7.5) regions on RNA, DNA, thymidine-5'-p-nitrophenyl phosphate, bis (p-nitrophenyl) phosphate, p-nitrophenyl phosphate and 5'-AMP. But as for polynucleotide phosphorylase the optimum pH was only in the acidic region (pH 5.5) on 5'-ADP.2) The optimum temperature was 60℃ for both acidic and alkaline RNases. They were considerably heat stable and 90℃ of their activities were retained after they were heated at 60℃ for 30 min. They were also stable at pH 5.0-7.5.3) These acidic and alkaline RNases were activated by Mg^<++>, Ca^<++> and Mn^<++> and their activities were inhibited by a few chelating and reducing agents.4) 5'-mononucleotides, 3'-mononucleotides and nucleoside-5'-diphosphates were contained in hydrolysates of RNA by the preheated crude enzymes from these sprouts. Especially, a large amount of nucleoside-5'-diphosphates was produced by enzymes obtained from barley roots.5) In the sections of millet sprouts, the distribution of nuclease was characterized as follows; 5'-nuclease comparatively specific to pyrimidine base existed in the root, while 3'-nuclease specific to pyrimidine base existed in the cotyledon and endosperm.
