In previous papers, we have described the properties of cellulolytic enzymes and the isolation of carboxymethylcellulase from filtrates of Aspergillus Saitoi.The present paper is concerned with the effects of inhibitors and activators of the purified carboxymethylcellulase.Of the metal ions tested, Hg^<++>, Mn^<++++>, pb^<++>, Ag^+, Mn^<++>, Ca^<++> and EDTA had inhibiting effects. Hg^<++> was the most effective followed by Pb^<++>, Mn^<++++>(each 10^<-2M>)which reduced enzyme activity at pH 4.0 approximately 50%. While the others(Mn^<++>, Ca^<++>, EDTA) had slightly inhibiting effects. On the other hand, Cu^<++>)10^<-2M>)and Co^<++>(10^<-4M>)increased activity at pH 4.0 to 170% and 160% respectively.Thiol reagents (iodoacetic acid, p-chloromercuribenzoate) and reducing-oxidizing agents(potassium dichromate, potassium cyanide)were inhibitory. Iodoacetic acid and p-chloromercuribenzoate inhibited enzyme activity at pH 4.0 in a concentration of 10^<-4M> and 10^<-3M> to about 50% and 40% respectively, and also potassium dichromate reduced enzyme activity at pH 4.0 to about 50%.On the contrary, potassium ferricyanide activated enzyme activity to approximately 25% when presented in concentration of 10^<-3M> at pH 4.0.Furthermore, the effects of a number of dyes were also tested for inhibition and activation on enzyme activities. Methyl violet and bordeaux red had the greatest activating effects ; the former was about 270% and the latter about 180%. But the others, except that eosin fuchsin, benzopurpurin, congo red, methyl violet and bismark brown were slightly inhibitory, had no effects.The addition of protein to this enzyme increased slightly its activity and also the pre-incubation of enzyme in the presence of protein had a protective function for its activity.
