In previous papers, the properties of cellulolytic enzyme from Aspergillus Saitoi culture filtrates have been described by the authors, and an enzyme having a specific activity towards CMC-Na, carboxymethylcellulase has been purified to give an electrophoretically homogeneous protein, and some properties of this enzyme have been reported.This report concerns cellulolytic enzyme production of Asp. Saitoi strain Sakaguchi Iizuka et Yamazaki IFO 6670 under various conditions.Production of carboxymethylcellulase having an activity on CMC-NA was investigated using a submerged culture under several circumstances.(I) A rather higher corn steep liquor concentration as a nitrogen source rather than dextrin as a carbon source, was discovered to be reguired for the production of enzyme.Such a finding is opposite when compared with enzyme production of most fungi.(II) The effect of varying ammonium phosphate concentrations was tested and it was found that the addition of 1 percent resulted in a better harvest than 3 percent.(III) Calcium carbonate bad influence upon enzyme production, but not on growth, because it interfered with pH lowering in the course of fermentation. Also the culture was found to have increased in cellulase activity in the presence of 3 percent ammonium sulfate, even though growth was delayed.(IV) The addition of a substrate, such as CMC-Na or cellulose powder, caused a slight increase or stimulation in cellulase production.(V) pH good cellulase production in culture medium was obtained with pH 3.1.(VI) Cellulase activity appeared in the culture at about the time that pH was at its lowest point in a range from 3 to 4.A study was also made of cellulolytic enzyme fermentation using a tank culture procedure according to the results of the submerged culture study.Generally, cellulolytic enzymes rapidly appeared in the medium when the initial pH was low, but the comsumption of carbohydrate by fungi was delayed and the mycelium weight did not increase much. Regarding cellulolytic enzyme production, it was recognized that enzymes (C_1) which degrades cellulose powder, CMCase (Cx) and β-glucosidase all of which were produced successively during the fermentation of Asp. Saitoi, had optimum initial pH in the medium for cellulolytic enzyme production of about 4-5,3-4 and 3 respectively.
