In general, native cellulose is resistant to enzymatic hydrolysis than swollen or other modified celluloses. In this study, enzymatic degradation of wood pulps, cotton linters and filter papers was investigated using crude cellulase preparations of Trichoderma koningi. Breakdown of filter papers was investigated by means of Toyama's method, estimation of reducing sugar, amount of residual cellulose, degree of polimelization and microscopic observation. The filter papers tested in this study was prepared from cotton linter which had received different degree of beating treatment. Thus, the following was recognized. Concerning the reducing sugar production, wood pulps and well beaten filter paper were decomposed about 90% of their original amount in 120 hours. However, in respect of residual amount, native cotton linters were hydrolyzed only 22% of their original amount by the same treatment. Cotton linters which received alkali treatment and bleaching treatment turned to be more hydrolysable than the native linter. Degree of beating treatment affected remarkably on the filter paper breakdown assay value. In our study, the filter paper which was made from seven hours beaten cotton linter needed minimum time to breakdown. The quality of filter paper should be strictly controlled in the filter paper breakdown assay of cellulase. Some discussion was noted and discussed.
