Comparison was made among three kinds of methods to find out a conventional assay method for cellulolytic activity. The enzyme preparation used here was obtained by adding alcohol as precipitant to the extract from wheat bran culture of Trichoderma koningi which was furnished by Dr. N. Toyama, Miyazaki University.Three methods i.e., 1) cup assay method, 2) estimation of liquefying activity for CMG and 3) estimation of breakdrown of filter paper, were studied. Reffering the latter two methods, analysis of reducing sugar produced was also made.Then, the correlation amoung the estimates by these three methods were inquired. It was unable to find out any correlation between 1) and 2), or 1) and 3), however, correlation was observed between 2) and 3).Five kinds of commercial cellulase preparation derived from Aspergillus niger ware compared with each other by means of 6 kinds of different assay methods. These preparations showed different activity patterns.Thus, the authors arrived at a conclusion that the estimation of cellulase activity should be made by means of two or more different assay methods, because commercial cellulase preparation might generally contain several kinds of cellulases.
