This paper describes a method which facilitates the isolation of the auxotrophic mutants of fungi. An example is given as follows.A suspension in the minimal medium of Aspergillus niger conidia is irradiated by ultraviolet light until the survial rate becomes 5% of the untreated control. After incubation for 15〜24 hrs., the suspension is filtered through the glass filter No. 3 in order to remove the mycelia which have been grown. Then, a portion of the filtrate previously diluted with sterile water is spread over the plates of the complete medium so at yield 20〜30 colonies per plate by incubating for 2 dyas at 30℃. After that, the temperature is lowered to 20℃, at which it is incubated further for 1〜2 days to give rise to sporulation. Then, conidia of colonies formed are transferred to the minimal medium plates by platinum needls manually and the plates are incubated at 30℃ for 1 day while prototophic strains yield distinct colonies. The spots of conidia which remain ungerminated are then transferred to the slants of complete medium to obtain cultures of auxotrophic mutants. The method saves the time, labour and media for isolation.
