1. Agarase of Vibrio agarliquefaciens FUKUMOTO et ISHIMATSU is an adaptative enzyme, and about 0.5% agar is suitable for its production.2. Despite the presence of agar agarase production is greatly inhibited by the addition of sugar to the media.3. For the secretion of agarase, peptone and asparagine are the most suitable nitrogen sources, and some inorganic nitrogen sources such as (NH_4)_8PO_4,(NH_4)_2HPO_4,KNO_3 and (NH_4)_2SO_4 are also effective, but NH_4Cl is inadequate.Bouillon containing agar has the same effect as the synthetic media described above for the production of agarase, but alkaline extract of soy bean cake does not so affect, even in the presence of agar.4. The concentration of peptone in 0.15〜0.7% or of KNO_3 in 0.1〜0.5% exerts no remarkable influence on the agarase production.5. The agarase production in the culture solution reached to its maximum after 5〜7 days, but any distinct relation between the incubation period and the enzyme secretion was not recognized.6. Addition of ethyl alcohol or acid bydrolysate of sardine meal(raw amino acids mixture) to media brings no increase in the agarase production.
