(1) In the search for amylase producing bacteria, an aerobic, spore forming, non-motile bacillus was isolated from soil, which possesses high amylolytic activity. It belongs to Bacillus alhaerens group in Bergey's manual, and resembles to Bacillus panis Migula, but it is different from it by following points : Spores terminal, thick scum formation, non-turbidity in broth, no reduction of nitrate, strong starch liquefication and optimum temperature (37℃). Moreover, it does not produce H_2S in broth and ferments no carbohydrate and its spores endure 8 hour's heating in boiling water.Therefore, I designate this species Bacillus Morigucliensis n. sp.(2) The fundamental conditions of amylase production are 30℃, pH 5.5,and 4〜5 days' culture.(3) The results of preliminary tests on surface and submerged process were as follows.On surface process with the media prepared by extraction of 5% defatted soy-beans, the activity of amylase became as high as 750 Lintner-Sollied value (L.S.V.) per 1 ml of solution, within 4 days at 30.C.On submerged process using shake-flask, the production of amylase is more efficient. hen Wthe media containing 2% wheat-bran and 3% defatted soy-beans were used, the value of 1,000 L.S.V. per 1 ml was obtained within 70 hours at 25℃.
