Breeding by Interspecific Protoplast Fusion of Koji-Molds, Aspergillus awamori var. kawachi and Aspergillus usamii mut. shirousamii

A new breeding technique for Aspergillus awamori var. kawachi and Aspergillus usamii mut. shirousamii involving interspecific protoplast fusion was used to develop new koji-molds for making spirits for drinking. A strain obtained from a heterokaryon strain using d-camphor showed 1.5-1.7-fold DNA content per conidium and about 1.3-fold conidial diameter compared to those of A. awamori var. kawachi and A. usamii mut. shirousamii. This strain was regarded as a heterozygous diploid. The strain was treated with benomyl as a haploidizing agent, thus obtaining many segregants. The prototrophic segregants were selected and their haploidy and diploidy were confirmed by measurement of the conidial size and DNA content per conidium. From these results, some segregants were considered to be haploid recombinants. A prototrophic segregant showed higher citric acid productivities than that of A. awomori var. kawachi and A. usamii mut. shirousamii.
