A native cellulose hydrolyzing cellulase was isolated and purified by Sephadex G-75 gel filtration and DEAE-Sephadex A-50 column chromatography from a submerged culture of Trichoderma resei QM 9414. The cellulase component showed a single protein band by polyacrylamide gel electrophoresis. The molecular weight of the cellulase was estimated to be 50,000. The cellulase hydrolyzed more specifically water-isoluble cellulose such as absorbent cotton and Avicel than water-soluble cellulose such as carboxy-methyl-cellulose. The cellulase was an essential cellulolytic component of T. reesei QM 9414. The cellulase produced glucose, cellobiose and cellotriose from cellotetraose, cellopentaose and cellodextrin and we concluded that the cellulase attacked cellulose randomyl. Degradation of absorbent cotton and tissue paper by the native cellulose hydrolyzing cellulase was microscopically observed.
