Screening and Identification of a Cyclomaltodextrin Glucanotransferase-Producing Bacteria

Fourteen strains of bacteria were screened for cyclomaltodextrin glucanotransferase-producing activity. Based on dextrinizing assay and cyclodextrin formation, C31 was chosen as the best enzyme-producing strain. The rod-shaped, sporeforming, and aerobic characters of C31 clearly indicate the genus Bacillus for this bacterium. In the species identification, C31 mostly resembled B. circulans in its cultural and biochemical characteristics. Genetic analysis through the G+C content also reveals that C31 could be reasonably well-fitted in the very heterogeneous group of B. circulans. It was found that C31 produces β-cyclodextrin as a predominant product of the degradation of starch through the action of cyclomaltodextrin glucanotransferase. In addition, the ratio of β- to α-cyclodextrins formed was also reported.
