Isolation and Properties of Ethanol-Using Yeasts with Acid and Ethanol Tolerance

Ethanol-using yeasts, strains IM-10,WY-1,-2,-3,-4,and NK-4,were isolated from natural sources after enrichment culture in a medium (pH 3.0) containing 8% (v/v) ethanol as a sole carbon source.Strain IM-10 was identified as Issatchenkia sp. and strains WY-1,-2,-3,-4,and NK-4 were Candida sp., respectively, on the basis of its morphological and biological characteristics, coenzyme Q system and GC content.Optimum conditions for growth of these strains were at pH 3-4 and 30-33℃, and in the presence of 1-3% ethanol. NK-4 grew well at pH 2.0.Lyophilized cells of the isolates contained 33-40% crude protein, 29-37% carbohydrate and 6-10% nucleic acid.Strain WY-1 used about 50% of ethanol added when incubated at 37℃ for 24 h in a medium (pH 2.5) containing 5% (v/v) ethanol.
