Chrogalin, a Heat-Stable Glycoprotein That Inhibits Polygalacturonase : Isolation, Characterization and Biological Activity

A glycoprotein that specifically inhibits polygalacturonase activity, named chrogalin, was isolated from culture broth of a Pezizales by precipitation with acetone and gel filtration in Sepharose 6B. Chrogalin had a molecular weight of 〜10^6 and contained 8.9% carbohydrate which was composed of mannose, glucosamine, galactose, glucose and arabionose (47 : 40 : 5 : 4 : 4). Chrogalin was stable at 100℃ at pH 5-9. Polygalacturonases of microbial orgins were inhibited 50% at a concentration of 10μg/ml of chrogalin. Maceration of tissue slices of different plants by fungal polygalacturonase was significantly reduced by chrogalin at 0.4-1 mg/ml.
