Factors Responsible for Liberation of β-N-Acetylglucosaminidase Bound to Mycelia of Streptomyces sp.

Streptomyces L-13 strain produced extracellular and mycelium-bound β-N-acetylglucosaminidases. Most of the enzyme was of the bound type when the strain was grown on a medium composed of glucose, yeast extract and inorgenic salts. The bound enzyme was most readily liberated by incubation of mycelia with 0.3 M phosphate buffer of pH 6.5 at 40℃ for 1 h, but even under optical conditions 50 to 80 percent of the enzyme was retained on the mycelia. The retained enzyme was further released by trypsin. The liverated enzyme was bound again to the mycelium by addition of an inorganic Mg^<2+> salt to the medium.Replacement of yeast extract in the medium by casamino acid accelerated accumulation of the extracellular enzyme. The mycelia cultivated in this medium also had mycelium-bound protease, which was produced only in small amount in the glucose-yeast extract medium. An increase in the amount of extracellular β-N-acetylglucosaminidase was accompanied by an increase in that of bound protease. The optimal pH for the protease activity was similar to that for the release of β-N-acetylglucosaminidase from the same mycelium, i.e., 6.5. An extract prepared from mycelia actively secreting β-N-acetylglucosaminidase contained protease activity and could release β-N-acetylglucosaminidase from the mycelia grown on the glucose-yeast extract medium.
