Diffusional and Electrostatic Effects on the Kinetics of Immobilized Enzymes

An expression for the overall effectiveness factor incorporating the combined diffusional and electrostatic effects for an immobilized enzyme catalysed reaction has been proposed in order to quantitatively evaluate the effect of partitioning of a charged substrate in and outside the carrier on the overall reaction rate.Numerical results for the overall effectiveness factor were computed for both the case of a small pore size, where the thickness of the electrostatic double layer is of the order of the pore size, and for that of lrge pore size, where the pore size is bigger compared to the electrostatic double layer. The effect of external mass transfer coefficient and the electrostatic potential on the overall reaction rate were also evaluated. The significance of the partitioning coefficient on the proper selection of a carrier for enzyme immobilization is stressed.
