Influence of Culture and Nutritional Conditions on the Production of Lipase by Submerged Culture of Aspergillus niger

Studies of submerged culture revealed that the culture and the nutritional requirements of Aspergillus niger E.I. 202 differed from those of previously reported strains. The optimum pH, temperature and incubation period were 7.0,35℃ and 96 hr, respectively. Olive oil along with sucrose (1%) was the best carbon source. Na^+, K^+, Ca^<++>, Cl^-, SO_4^<--> had no effect on lipase synthesis. Mg^<++> had good effect on the mycelial growth but had an inhibitory effect on lipase synthesis per unit weight of mycelia. Fe^<++> plays a very important role in the synthesis of lipase. Fe^<++> in combination with Mg^<++> gave good growth and the maximum yield of lipase. A medium containing NH_4NO_3 (0.1%), KH_2PO_4 (0.2%, MgSO_4・7H_2O (0.04%), FeSO_4・7H_O (0.1%), sucrose (1%) and olive oil (1%) was suitable for optimum yield of lipase.
