Report 14 and 15 informed that acceleration of tuberculin reaction by repeatedly injected site is specific to tuberculin. Tuberculin reaction is a representative delayed type reaction, caused by the protein fraction of tuber-culin. Is the early reaction at the repeated site also caused by yhe protein fraction or by the polysac-charide fraction? In this report, the influence of the injection of tuberculin fractions on the susbequent tuberculin reaction at the same site was investigated: 1) Old tuberculin was injected in two new sites on the backs adults. After one or two months, 0.06mcg of PPD and 0.06mcg of CF (polysaccharide fraction of old tuberculin) were injected in one of these two previously injected sites respectively, and in two new site as a control. Reactions were observed 4, 8, 24, 48 hours after injection. Early reaction was observed only at the site where at first tuberculin and secondly PPD were injected. At the site where at first tuberculin and secondly CF were injected, early reaction was not observed and the reaction pattern was almost the same at the new site. 2) 0.06mcg of PPD and 0.06mcg of CF were injected in two sites respectively on the backs of six adults. After one month old tuberculin was injected in two previously injected sites, where PPD or CF had previously been injected, and in a new site as a control. Reactions were observed as previous experiment. Early reaction was observed only at the site where at first PPD and secondly tuberculin were injected. At the site where at first CF and secondly tuberculin were injected, early reaction was not observed and the reaction pattern was almost the same at a new site. 3) 0.06mcg of PPD was injected in two sites new on the backs of seven adults, and 0.06mcg of CF was also injected in two other sites. After one month PPD was injected in two previously injected sites, where PPD or CF had previously been injected respectively, and in a new site as a control. CF was also injected in the remaining two previously injected sites and in a new site. Reactions were observed as previous experiment. Eearly reaction was observed only at the site where PPD was twice injected. At the site, where at first CF and secondly PPD were injected, reaction patterns were almost the same in a new site. At the site where CF was secondary injected neither early reaction not delayed reaction was noticed. 4) Old tuberculin was injected in two new sites on the backs of four adults. After one month tuber-culin and "tuberculin digested with trypsin" were injected in one of these two previously injected sites respectively, and in two new sites as a control. The observation times were the same as in the previous report. At the site where tuberculin was twice injected early reaction was observed in all four cases in contrast to the new site. At the site where at first tuberculin and secondly "digested tuberculin" were injected, early reaction was not observed and the reaction pattern was almost the same as at a new site. In summary, the results of these exepriments suggested that the tuberculin fraction responsible for the early reaction might be the protein fraction and not the polysaccharide fraction.
