1) Reaction of rhumatoid factor with immune precipitates: To the quantitative precipitation reaction with egg albumin and anti-egg albumin rabbit serum, fresh rheumatoid serum was added. There was a considerable increase in nitrogen value of the precipitates as compared with that of the control series. The similar results were obtained in the systems of human thyroglobulin and serum with Hashimoto's thyroiditis, and cytochrome C and anti-cytochrome C rabbit serum. This was most marled in the zone of slight antigen excess. Addition of inactivated rheumatoid serum, exhibited less increasing effect as compared with that of nonheated one. Normal human serum had no such effect. It would be reasonable to presume that rheumatoid factor reacts with human or rabbit antigen antibody complexes and absorbs complement. 2) Reaction of rheumatoid factor with immune hemagglutinates: The addition of rheumatoid serum caused marked increase in the agglutination titers of the hemagglutination reaction in the following systems (7 Systems). #1) Isohemagglutination (blood group A) #2) Staphylococcal hemagglutination #3) Sheep cell hemagglutination #4) Penicillin-albumin tanned cell hemagglutination On the contrary, no significant increase of titers was observed in the following reactions (19 S system. ) #5) Penicillin sensitized hemagglutination #6) Human cold hemagglutination These results support the view that the rheumatoid factor may be an antibody against antigen antibody complexes (i. e. aggregated 7 S gamma glubulins). 3) Absorption of rheumatoid factor with immune hemagglutinates: Hemagglutinates based on next three forms of antigena-ntibody binding were used in this study. #1) Thyroglobulin and serum with Hashimoto's thyroiditis #2) Penicillin-albumin conjugate and rabbit anti-penicillin serum #3) Sheep cells and rabbit anti-sheep cell agglutinin The rheumatoid factor activity was relatively easily absorbed with the hemagglutinate produced in the system #1. No absorption of rheumatoid factor was seen in the rabbit antigen antibody systems (#2 & #3).
