The circulating antibody present in primed animals combines with secondarily administered antigen and resulting complexes are rapidly removed from circulation by the reticuloendothelial cells. In the present study, it was clarified that this mechanism is important on the antigen recognition in secondary immune response. When the spleen or lymph node cells (NSC or NLC) from non-immunized mice were exposed with bacterial α-amylase (BαA) complexed by mouse anti-BαA antibody, in vivo or in vitro, thoroughly washed, then mixed with memory cells and transferred to X-irradiated recipient, marked secondary anti-BαA antibody production was observed. From the experiments using NLC passing through glass wool column or thioglycollate-induced peritoneal macrophage, it was suggested that the macrophage play a role in the antigen recognition of secondary response, and that the immunogenicity of antigen was facillitated by the enhanced ingestion of antigen in the presence of antibody. The Fc portion of antibody is important, but cytophilic antibody and complement may not participate in this process. From these results, the possible mechanism of antigen recognition was discussed.
